As memórias, os pensamentos e o comportamento são codificados por alterações na força da comunicação sináptica. Estas alterações constituem a base celular e molecular do processamento de informação subjacente ao funcionamento do cérebro. No entanto, as transformações funcionais utilizadas pelas sinapses para codificar e processar a informação são ainda pouco conhecidas. Estamos interessados nos mecanismos celulares e moleculares da excitabilidade neuronal, da função sináptica e da plasticidade, e na forma como a disfunção sináptica está subjacente às doenças neuropsiquiátricas e neuroimunes.

Utilizamos uma combinação de técnicas como culturas primárias de neurónios dissociados e cortes cerebrais, bioquímica, biologia molecular e celular, genética molecular de ratinhos, eletrofisiologia e análise comportamental para estudar o papel dos intervenientes moleculares e dos processos neuromoduladores que regulam os receptores de neurotransmissores, a função e plasticidade sinápticas.

Esta investigação fundamental tem fortes implicações para as perturbações cognitivas, uma vez que as variantes genéticas em múltiplas proteínas sinápticas estão ligadas à deficiência intelectual, à esquizofrenia e às perturbações do espetro do autismo, e para as perturbações neurodegenerativas associadas à idade estão na sua fase inicial originadas por defeitos na transmissão sináptica. Concentramo-nos nas alterações da função sináptica relacionadas com a doença, quer sejam genéticas, desencadeadas pela experiência ou por auto-anticorpos que visam proteínas do sistema nervoso central, para compreender como a disfunção sináptica está na base da patogénese da doença. Os nossos estudos celulares e moleculares e os modelos animais que estamos a gerar podem também contribuir para o desenvolvimento racional de terapias para estas doenças.

Focos de investigação:

  • Investigar a coordenação da plasticidade sináptica e intrínseca e a sua desregulação nas perturbações do neurodesenvolvimento. PI: Ana Luisa Carvalho
  • Compreender a resposta sináptica ao stress crónico, incluindo em condições como a depressão e a COVID-19 prolongada. PIs: Paulo Pinheiro, Ester Coutinho e Ana Luisa Carvalho
  • Compreender como a disrupção imunomediada ou genética de proteínas sinápticas (e.g. CASPR2, SNAP29, stargazin,) contribui para défices no desenvolvimento neuronal e na função sináptica em doenças do neurodesenvolvimento, usando modelos animais e culturas neuronais derivadas de humanos geneticamente editadas.   • Elucidar a forma como os auto-anticorpos dirigidos a proteínas sinápticas são patogénicos; conduzir um estudo nacional multicêntrico centrado na encefalite autoimune seronegativa com o objetivo de identificar novos alvos antigénicos; estabelecer uma rede de colaboração entre clínicos e cientistas portugueses com conhecimentos especializados em encefalite autoimune.
