Uma função cerebral adequada depende do equilíbrio entre sinapses excitatórias e inibitórias, as quais são reguladas por sistemas neuro-moduladores. O trabalho desenvolvido por este grupo é inspirado pelo impacto da cafeína, o principal componente do café, no aumento do estado de atenção, na normalização da memória e do humor, no aumento da esperança de vida e envelhecimento ativo e na sua capacidade de impedir o aparecimento de diferentes distúrbios neurodegenerativos (doenças de Alzheimer e de Parkinson).Assim, o objetivo do grupo é estudar os sistemas de neuro-modulação operados por purinas (adenosina e ATP), conhecidas por serem alvo da cafeína, para dessa forma interferir na evolução de doenças neurodegenerativas. Identificámos os recetores A2A da adenosina (A2AR) como os principais alvos da cafeína no controlo de distúrbios cerebrais, usando uma combinação de ferramentas farmacológicas, genéticas e opto-genéticas. Estabelecemos que os A2AR são necessários e suficientes para desencadear sintomas em diferentes modelos animais de doenças de Alzheimer ou Parkinson, depressão, epilepsia ou encefalopatia diabética, por meio de uma capacidade combinada de restaurar a plasticidade sináptica, controlar a neuroinflamação e reforçar o suporte metabólico pelos astrócitos.
O grupo tem diferentes animais (murganhos) transgénicos e modelos de doenças cerebrais induzidas farmacologicamente, bem como um conjunto de ferramentas para interferir com o A2AR (murganhos transgénicos “cel Specific” e sistemas mediados por vírus para reforçar ou diminuir a função dos A2AR) e acesso a tecido cerebral humano.
Seguimos uma abordagem holística das questões científicas, agregando a nossa experiência em: i) estudos de comportamento animal que avaliam aprendizagem e memória, desempenho motor, alterações de humor e processamento do medo; ii) cirurgia cerebral estereotáxica; iii) estudos eletrofisiológicos ex vivo (fatias do cérebro) para avaliar a função de circuitos cerebrais (registros extracelulares) e correntes iónicas (patch-clamp); iv) imagiologia de alterações funcionais (“cell live Ca2+”), morfológicas (histoquímica) e neuroquímicas (ligação, tráfico e expressão de receptotres), combinando técnicas de radioligandos, FRET, ensaio de ligação por proximidade, PCR e de Western blot); v) fracionamento sub-celular de sinaptossomas e gliossomas e uso de culturas celulares (neurónios e astrócitos) para estudar a liberação e captação de neurotransmissores.
O grupo tem um histórico de mais de 250 publicações em revistas científicas com alto fator de impacto, com mais de 12.000 citações e um fator h de 62, com base em inúmeras colaborações em todo o mundo. O grupo tem uma vasta experiência em treino de pós-graduação (mais de 15 estudantes de pós-doutoramento, 40 de doutoramento e 25 de mestrado), com financiamentos obtidos de agências americanas e europeias, bem como de empresas privadas e farmacêuticas.
